Protein-carbohydrate interaction. V. Further inhibition studies directed toward defining the stereochemical requirements of the reactive sites of concanavalin A.

چکیده
Inhibition studies on the concanavalin A-dextran interaction have been extended using as inhibitors the members of the maltose-, isomaltose-, and methyl cy-isomaltoside series, amino and N-acetyl amino sugars, and a number of oligosaccharides containing more than one linkage type. The interaction of the plant protein at the terminal chain ends of the a-linked glucans and mannans has been essentially confirmed, and the different characteristic inhibition activities of the linkage types is discussed in terms of steric effects. A possible application of the inhibition technique towards structural analysis of carbohydrate molecules is suggested by the authors.

Protein-Carbohydrate Interaction III. Agar Gel-Diffusion Studies on the Interaction of Concanavalin A, a Lectin Isolated from Jack Bean, with Polysaccharidesl
Concanavalin A, a protein isolated from the jack bean (Canavalia ensijormis), has previously been shown to form a precipitate with glycogen and yeast mannan, and more recently with dextrans and amylopectin. Double-diffusion precipitation studies in agar gel have been found to afford a very sensitive and satisfactory method for investigating the interaction of concanavalin A with certain types o...
متن کاملQuantitative studies on the interaction of concanavalin A, the carbohydrate-binding protein of the jack bean, with model carbohydrate-protein conjugates.
A ~ t T h e interaction of concanavalin A with a comprehensive series of carbohydrate-bovine serum albumin conjugates was investigated by agar gel diffusion and quantitative precipitation techniques. These studies showed that when conjugated to bovine serum albumin, those sugars which inhibited concanavalin A-polysaccharide interaction formed a precipitate with concanavalin A. The unexpected re...

The immunochemical techniques of quantitative precipitation and hapten inhibition have been employed in a study of the interaction of concanavalin A with several.levans of bacterial and plant origin. Of the polysaccharides that form a precipitate with concanavalin A (dextrans, D-mannans, glycogens, amylopcctins, and levans), levans appear to be the least reactive. Hapten-inhibition studies were...
متن کاملDirected Blocking of TGF-β Receptor I Binding Site Using Tailored Peptide Segments to Inhibit its Signaling Pathway
Background: TGF-β isoforms play crucial roles in diverse cellular processes. Therefore, targeting and inhibiting TGF-β signaling pathway provides a potential therapeutic opportunity. TGF-β isoforms bind and bring the receptors (TβRII and TβRI) together to form a signaling complex in an ordered manner. Objectives: Herein, an antagonistic variant of TGF-β (AnTβ)...
متن کاملEffect of Tofu Wastewater Addition on the Growth and Carbohydrate-Protein-Lipid Content of Spirulina platensis (RESEARCH NOTE)
This study was conducted to investigate the effect of tofu wastewater (TW) addition to the growth medium on the growth of Spirulina platensis. The TW addition was varied in range of 0 – 8 %v/v. The results showed that the growth rate (μ) of S. platensis at TW addition of 0, 2, 4, 6, 8 %v/v was 0.007, 0.084, 0.074, 0.088, 0.086 mg/L/d, respectively. The medium with TW content of 6 %v/v (carbon:n...
